ACE
It is a member of the M2 gluzincin family of zinc metallopeptidases and functions primarily as a dipeptidyl carboxypeptidase, catalysing the hydrolysis of carboxy-terminal dipeptides from a wide range of functional peptides. ACE consists of two isoforms, somatic ACE (sACE) and testis ACE (tACE).The renin-angiotensin system (RAS) is a complex enzymatic pathway that plays a critical role in blood pressure regulation and generates a number of bio-active peptides which control fluid and electrolyte homeostasis, hormone secretion as well as behavioural and cognitive responses. ACE has long been established as having an important role in the RAS, with ACE inhibitors being a major component of the medical treatment of hypertension. Upstream of ACE, renin is a key regulator of the RAS. This enzyme is produced from the juxtaglomerular complex in the renal nephron in response to stress, decreased sodium levels or effective plasma volumes.
